John Buscema se sintió atraído por las historietas siendo niño, y copiaba las viñetas de Alex Raymond, Hal Foster, Burne Hogarth y Elzie Segar. Guió sus primeros pasos hacia la ilustración comercial dada de su evidente capacidad para el dibujo realista de la figura humana tras dejarse influenciar por los clásicos (los europeos Rubens, Monet, Renoir, Rafael, y los yanquis Al Dorne, Dean Cornwell, Norman Rockwell y Robert Fawcett). Su primer encargo profesional consistió en la realización de portadas para revistas militares, publicidad y algunas viñetas humorísticas que realizó para la publicación Hobo News. Creyó oportuno dibujar cómics luego, en un momento en el que la industria era boyante. Logró un puesto fijo en Timely (luego Atlas, luego Marvel) a finales de la década de 1940, realizando historietas de todo tipo de géneros, pero sobre todo westerns, románticas y policíacas. La primera historieta firmada por Buscema, “They Had Robbery Lincoln’s Grave”, llevó guión de Stan Lee. Su estilo rudo en un comienzo fue suavizándose y perfeccionándose a su paso por otros sellos editores (Quality, St. John, Ziff Davis o Charlton) y con la influencia de otros profesionales como Carl Burgos, Syd Shores o Gene Colan, siempre presididos por el gran Foster. Con este bagaje, Buscema se sintió poderosamente atraído por el dibujo de historia de corte épico e incluso fantástico en las que la protagonista era la aventura; una prueba de ello fueron sus adaptaciones para Four Color Series II (Dell Publishing).

Debido a la crisis padecida por la industria del cómic estadounidense durante los años cincuenta, Buscema volvió a la publicidad en 1958. Stan Lee le reclamó para los cómics de nuevo en 1966, y entonces inició su ya histórica carrera trabajando en el género de los superhéroes desde que sustituyó a Jack Kirby en una historieta protagonizada por Nick Fury. Así, durante los años sesenta cosechó fama con sus trabajos en Incredible Hulk y Namor the Sub-Mariner, gran consideración en Silver Surfer, y gloria en Fantastic Four, The Mighty Thor y The Avengers¸ hasta igualar la popularidad de Kirby. No obstante, Buscema sería identificado sobre todo con un héroe sin superpoderes: Conan, en cuyas series Conan the Barbarian, desde 1973, y The Savage Sword of Conan, desde 1974, consiguió sus máximos logros (al tiempo que también trabajaba sobre otros personajes de fantasía heroica en series como Kull the Conqueror, Red Sonja, King Conan). El llamado “canon Conan” fue el definido por Buscema y sus entintadores, generalmente de origen filipino, que con el paso de los años pasaría al imaginario popular y sería la base para la definición de la imagen cinematográfica del personaje, la más conocida.

Entre 1970 y 1990 Buscema fue un modelo a seguir (en su estilo gráfico, junto a John Romita) entre los dibujantes de la editorial Marvel y se convirtió en el perfecto ejemplo de autor prolífico e incansable, llegando a dibujar 60 páginas mensuales y llegando a publicar en aproximadamente 60 títulos distintos de Marvel (destacando su trabajo en The Avengers). Con todo, tuvo tiempo para impartir clases y para ilustrar libros didácticos. De nuevo popular a finales de los años ochenta gracias a su labor con el mutante llamado Wolverine, Buscema decidió retirarse en 1996, después de mostrar desgana en algunas de sus producciones últimas (Mighty Morphin Power Rangers, The Savage Sword, Conan the Savage). No obstante, un par de años después de esta jubilación -y tras haber denostado el género superheroico públicamente-, Buscema volvió a Marvel para dibujar de un modo sorprendentemente vigoroso pese a su edad, tanto superhéroes (Thor, Galactus The Devourer) como bárbaros (Conan the Barbarian. Death Covered in Gold). Su último trabajo en el medio fue sobre guión de Stan Lee –el mismo con quien comenzó su carrera allá por 1948– pero para una serie y una editorial con los que apenas si trabajó a lo largo de su trayectoria profesional: Superman, de DC. Asimismo, se hallaba ultimando otro proyecto, junto a Alex Ross.

De su obra pictórica, que siempre se resistió a exponer pecando de humildad excesiva, existe una pequeña muestra en el libro de Vanguard Productions The John Buscema Sketchbook.
